Iambic Therapeutics is a San Diego, California AI-driven drug discovery company founded in 2019 by Thomas Miller (CEO) and Fred Manby (CTO) that designs small-molecule therapeutics for oncology and other indications. Iambic builds proprietary AI models for drug discovery, including Enchant, a multimodal transformer that predicts clinical properties (pharmacokinetics, cellular and biochemical activity) of candidate molecules from early discovery data, and NeuralPLexer (now NeuralPLexer3), a flow-based generative model for protein-ligand complex structure prediction used to identify cryptic pockets and guide structure-based design. Both lines of work have been published at venues including NeurIPS 2025. The company's internal pipeline is oncology-led: IAM1363, a brain-penetrant selective HER2 / HER2-mutant inhibitor, completed Phase 1/1b dose escalation in 2025 and is in dose optimization with clinical activity reported at ESMO 2025; IAM-K1 (KIF18A) and IAM-C1 (CDK2/4) are preclinical, alongside undisclosed neurology, gastrointestinal, and inflammation discovery programs. In February 2026 Iambic announced a discovery and technology collaboration with Takeda worth up to ~$1.7B plus royalties, following a $100M+ private financing in November 2025 and total disclosed funding of approximately $334M. The company has been named to the CNBC Disruptor 50 list in 2025 and 2026 and has signaled intent to access public markets, including via a SPAC. Iambic's public GitHub footprint is limited to np-bench, a BSD-3 benchmark library for 3D biomolecular structure prediction; the underlying NeuralPLexer and Enchant models are proprietary and there is no public developer API, SDK, or commercial platform offering.
